MerR family of transcription factors comprises the large and diverse group of transcriptional regulators that may act as repressors and activators and control various metabolic processes in the cell. Based on the nature of their effector molecules and the architecture of their ligand binding domains, MerR-family regulators are subdivided into two groups. The first group is composed of heavy metal-sensing regulators (including the founding member of the family, MerR) and oxidative stress-sensing regulators (such as SoxR) [20230832]. The second group includes proteins that have larger C-terminal effector binding domains and recognize various aromatic and organic compounds that are often toxic [20230832].

Binding sites for the activators (and some repressors) of MerR family are localized in an extended spacer between canonical -10 and -35 promoter elements [21954605]. MerR-family regulators have a characteristic N-terminal HTH DNA binding domain (PF00376) and a variable C-terminal region that recognizes a particular environmental stimulus [20233934].
