Fur family of transcription factors includes metal ion-dependent regulators that maintain the iron, manganese, zinc and nickel homeostasis (Fur, Mur, Zur, Nur), or control the oxidative stress response in Bacteria (PerR) [17216355, 11282473, 18259067]. The Fur-family regulators are activated by metal binding to two distinct binding sites. The histidine-rich motif is involved in the cellular response to iron, while the four conserved cysteine motif binds zinc. The metal bound regulators form tight complexes with the promoter regions of their target genes, thus repressing or activating their expression, depending on location of the binding sites.

Fur-family regulators contain a characteristic domain (PF01475), which contains an N-terminal HTH DNA binding motif and C-terminal part required for metal ion binding and dimerization [12581348]. Fur-family regulators were found both in Bacteria and Archaea.
