Transcription factor NrdR negatively controls expression of genes for deoxyribonucleotide reductases (nrd) of all three types in most lineages of Bacteria. Novel DNA-binding motif (NrdR-box) were first discovered by application of the comparative genomic approach to the nrd genes in various taxonomic lineages of bacteria [15949864]. The NrdR transcription factor (previously known as hypothetical protein YbaD in E. coli) was attributed to the identified NrdR-box motif by correlating their occurrence with the phylogenetic distribution of ortholog families. NrdR proteins constitute a novel and unique family of transcription factors containing Zn-ribbon and ATP-cone motifs.

The DNA-binding motifs of NrdR showed significant conservation across diverse bacterial lineages. The predicted regulatory role of NrdR was finally confirmed in experiments conducted in Streptomyces spp. and in E.coli.
