Crp family of transcriptional regulators includes numerous characterized activators/repressors of diverse biological processes. The Crp subfamily of regulators in bacteria senses the cellular concentration of cyclic AMP (cAMP), a second messenger important in many biological processes. In E.coli and other enterobacteria, the cAMP-Crp complex acts as a catabolite activator protein that activates the expression of genes involved in the utilization of alternative carbon sources in response to glucose starvation [16547048]. In several other groups of bacteria (Shewanella, Xanthomonas, Pseudomonas), Crp is not required for catabolite repression but controls anaerobic respiration, amino acid metabolism, virulence and pathogenicity [22081386, 21345179, 12775705].

Another well-characterized subfamily of regulators in the Crp family is Fnr, anaerobic regulators that were characterized in E.coli and other proteobacteria, as well as in B. subtilis. Fnr regulators respond to the changes in redox state (e.g., the decreasing availability of oxygen), resulting in transcriptional activation of genes involved in anaerobic respiration [16547048]. Apart from these subfamilies, the Crp family includes several regulators that use other effector molecules (e.g. carbon monoxide, nitric oxide, 2-oxoglutarate) and control other biological processes (e.g. denitrification, nitrosative stress, nitrogen metabolism) [16547048, 19415759]. Crp-family regulators contain an N-terminal effector sensing and dimerization domain (PF00027) and a C-terminal HTH DNA binding domain (PF00325) [19415759].
