BirA repressors control transcription of the biotin biosynthesis operon, and in the same time BirA proteins act as a biotin-protein ligase. Biotin is an essential coenzyme required for central cellular metabolism and its biosynthesis is tightly controlled [20142036, 12368242]. Transcription of the biotin operon in E.coli is derepressed under biotin starvation conditions, when BirA repressors remain largely monomeric and bio operator is unoccupied. When all available acceptor proteins are biotinylated, BirA bind its target DNA operator as a dimer and thus negatively regulate transcription of the biotin operon [20142036]. more
