HexR transcriptional factor from the RpiR family has been implicated in regulation of glucose metabolism genes in Pseudomonas putida. We used a comparative genomics approach to reconstruct transcriptional regulons controlled by HexR orthologs in Proteobacteria [21849503]. HexR regulates various genes from the central carbohydrate metabolism. The predicted in Shewanella oneidensis HexR regulon was experimentally validated by using in vitro and in vivo approaches. Binding of 2-keto-3-deoxy-6-phosphogluconate to HexR destroys the DNA-regulator complex formation. HexR has a dual mode of action on various target promoters - repression of genes involved in catabolic pathways and activation of gluconeogenic genes.
