BglG family of DNA-binding transcription factors constitutes activators of sugar uptake and catabolism genes in Gram-positive bacteria [10438772, 22014119]. BglG-family activators are induced by specific sugars that serve as substrates of cognate phosphotransferase systems (PTS). In contrast to most prokaryotic transcriptional regulators, BglG-family regulators do not bind effector molecules and their activity is controlled by phosphorylation and dephosphorylation. BglG-family activators contain one or two N-terminal DNA binding domains (PF08279 and/or PF05043), one or two central PTS regulation domains (PRD) (PF00874), and a C-terminal PTS EIIA-like domain (PF00359).

BglG-family regulators are activated by phosphorylation of certain histidyl residues in their PRD domains by PTS components. One PRD domain is being phosphorylated by the EIIB component of a cognate sugar-specific PTS system, whereas the second PRD domain is phosphorylated by a general PTS component, the HPr protein, allowing sensing of the catabolite status of the cell (i.e. the presence of extracellular glucose via the glucose-specific PTS). In the presence of an inducing sugar, and in the absence of glucose, a BglG-type regulator is active and can dimerize on the target promoters to activate the target gene expression.
