Transcription factor XylR from the ROK protein family negatively regulates the xylose utilization in Gram-positive bacteria. Regulators from the ROK family are composed of sugar kinase-like domain fused to a short DNA-binding HTH domain. The XylR regulon and its DNA-binding motif were experimentally investigated in Bacillus subtilis and Clostridium acetobutylicum, where it controls utilization of xylose and xylosides. XylR represses its target genes in the absence of xylose. Application of the comparative genomic approach allowed to reconstruct orthologous XylR regulons in several lineages from the Firmicutes phylum [11750820, 20406496].
