Transcription factor IscR negatively controls iron-sulfur (Fe-S) cluster synthesis genes in Proteobacteria. It was originally investigated in Escherichia coli as a repressor of the isc and suf operons involved in Fe-S synthesis. The IscR regulators belong to the Rrf2 protein family and contain a [2Fe-2S] cluster, which is required for repressor function as a sensor of the Fe-S status of the cell. The comparative genomic approach was used to identify candidate IscR-binding motifs within DNA regions protected by IscR during the DNAse I footprinting experiments in Enterobacteria. Then IscR regulon was reconstructed by comparative genomic approach in other taxonomic groups including gamma-proteobacteria (5 lineages), beta-proteobacteria [16677314], and alpha-proteobacteria [17173478].
